چکیده

Colistin resistance in Acinetobacter spp. is increasing, resulting potentially untreatable nosocomial infections. Plasmid-mediated colistin of particular concern due to its low fitness cost and potential transferability other bacterial strains species. This study investigated the mechanism a clinical nosocomialis isolate from Cape Town, South Africa. A colistin-resistant A. was identified blood culture 2017. PCR Illumina whole-genome sequencing (WGS) were performed identify genes mutations conferring colistin. Plasmid on an Oxford Nanopore platform. mcr functionality assessed by broth microdilution after cloning gene into pET-48b(+) expressing it SHuffle® T7 Escherichia coli curing plasmid using 62.5 mg/L acridine orange. The minimum inhibitory concentration (MIC) 16 mg/L. mcr-4.3 detected WGS. No previously described found 24 024-bp RepB (pCAC13a). Functionality studies showed that recombinant did not confer E. coli. However, pCAC13a restored susceptibility nosocomialis. We describe first detection plasmid-mediated encoding Recombinant expression coli, suggesting may be plasmid-dependent or species-specific.

Stereochemical Trajectories of a Two-Component Regulatory System PmrA/B in a Colistin-Resistant Acinetobacter baumannii Clinical Isolate

Background: There is limited information on the 3D prediction and modeling of the colistin resistance-associated proteins PmrA/B TCS in Acinetobacter baumannii. We aimed to evaluate the stereochemical structure and domain characterization of PmrA/B in an A. baumannii isolate resistant to high-level colistin, using bioinformatics tools. Methods: The species of the isolate and its susceptibility ...

Racial Disparities in Schooling: Evidence From Cape-Town, South Africa

Racial disparities in education in South Africa have been large and enduring post-apartheid. However, moving beyond simple descriptions of these disparities to a more detailed explanation has proven to be elusive. In this dissertation, I develop and estimate a dynamic model of schooling and labor supply of youths in South Africa, and use it to study several potential explanations for the racial...

Genetic Features of MCR-1-Producing Colistin-Resistant Escherichia coli Isolates in South Africa.

A series of colistin-resistant Escherichia coli clinical isolates was recovered from hospitalized and community patients in South Africa. Seven clonally unrelated isolates harbored the mcr-1 gene located on different plasmid backbones. Two distinct plasmids were fully sequenced, and identical 2,600-bp-long DNA sequences defining a mcr-1 cassette were identified. Promoter sequences responsible f...
